Can I roll a stally to 33y and then extract his semen?

Yes, you can.

But you do have to be careful not to roll him past 33. I usually extract about ten from my studs so I have back up if I accidentally roll into retirement. If I don't need them I just delete them and then they can be extracted again later.

I actually roll mine a few turns shy of 33 for the same reason, in case I accidentally turn them while moving them around. lol.

Semen is, well you know how you can put horses up for stud? And if a mare in heat breeds to it and gets pregnant, semen obviously has to be there. Studs can only be used up to 23 (?) and then you can extract semen, costing $20k per time. You can extract 20 vials. This means you can roll your horses for longer and thus getting better stats. So instead if clicking breed this stallion to this mare, or whatnot, you just use the semen. (I'm talking VHR terms)
